Pune does misal pav best, from what I've heard. Haven't tried it there so I can't confirm it.
I've done some research. Turns out my buddy was being biased in favour of his hometown. Apparently Pune tones down the spiciness. Misal Pav seems to have originated in Kolhapur and they have the spiciest versions of it. So I'd say it's either Kolhapur or Mumbai.

No one's ever going to Kolhapur, so it's Mumbai. Kolhapur one of those places you hear about all the time but don't know anyone who actually went there.

I've stopped there for food before. And yes, the Misal Pav really is amazing (at least at the restaurant I went to). Even the Vada Pav in the town was really spicy + they gave me a handful of green chillies to boot.

But I'm a spicehead so I loved every bite.

There is this village/small town called Ozhar nearby Nashik. I had tried Misal Paav there and it was just amazing as well.
